is filling of it return is mandatory for people who has income less than basic exemption limit from speculation activities?

Not mandatory but the loss can only be carried forward if the return has been filed.

If the exemption limit has not crossed then there is no requirement for filling of Return.

it is not necessary ti file IT return if total income of an assessee is less than basic exemption limit...
